What is the mode of action of Provera?

Medroxyprogesterone belongs to the class of drugs called progestin. It shows its remedial effect by binding to the to the progesterone receptors. After binding to the progesterone receptors, it reduces the liberation of Gonadotropin-releasing hormone from the hypothalamus that blocks the pre-ovulatory LH surge.

What is the dosing regimen of Provera?

Provera is commercially obtainable in an oral tablet form in dosing strengths of 5mg and 10mg. A woman with secondary amenorrhea should take 5 or 10 mg of Provera orally daily for 5 to 10 days. A woman suffering from abnormal uterine bleeding should take 5 or 10 mg of Provera orally once a day for 5 to 10 days; she should use this medicine from 16th or 21st day of the menstrual cycle. A postmenopausal woman relying on the treatment of conjugated estrogens should take 5 or 10 mg of Provera orally daily for 12 to 14 consecutive days per month to avoid the risk of Hyperplasia.

Who should not use Provera?  

The use of Provera is not safe for women who are suffering from any of these medical cases:

  • Undiagnosed unusual vaginal bleeding
  • Pregnancy
  • Breastfeeding condition
  • Breast cancer
  • A heart attack or stroke within the past 12 months
  • A previous record of hepatic impairment
  • A previous record of clotting disorder
